Sanitizer Testing

Purpose: The purpose of this lab is to test a new sanitizer for its ability to kill three species of bacteria.  The organisms to be tested are Staphyloccus aureus, Escherichia coli and Pseudomonas aeruginosa.  All three of these organisms can be found in hospital and medical facilities and pose a serious risk of infection for patients in these facilities.

Materials: Each pair of students will be given one organism to test (diluted overnight culture) along with the following
     1.  5 tubes of sterile dH2O - 9 ml per tube
     2.  Sterile 1 ml pipettes and pipettor 
     3.  Sterile nylon swabs
     4.  10 plates of TSA media

Protocol: Each pair of students will follow the protocol below:
     1.  Record name of your organism on your data collection sheet.
     2.  Label 5 tubes of sterile dH2O 1 through 5.
     3.  Preform a 1/10 serial dilution of the organism as directed by the instructor - transfer 1 ml of the organism to tube 1 and mix. Transfer i1 ml to tube 2 and       
          mix...repeat the process until you transfer 1 ml from tube 4 to tube 5.
     4.  Label 5 TSA plates "Control" and label each with a corresponding number of the dilutions; i.e. Control 1, Control 2, Control 3, etc...,the organism and the
          pair's initials.
     5.  Label 5 TSA plates "TEST" and label each with the corresponding number of the dilutions; i.e. TEST 1,TEST 2, TEST 3, etc..., the organism and the pair's
          initials.
     6.  Transfer 0.1 ml from the dilution tubes to the appropriate Control and Test plates; i.e. 0.1 ml from tube 1 to the plate labelled Control 1 and 0.1 ml to the
          plate labelled TEST 1.
     7.  Using a sterile swab spread the bacteria completely over the surface of the plate to create a lawn.
     8.  Once all of the plates have been inoculated, place the Control plates into the incubator and the instructor will give specific instructions on how to TEST will
          be tested in the sanitizer.
     9.  The TEST plates will be exposed in the sanitizer per the instructions provided by the manufacturer.  The exposure time was 30 seconds
     10. Once the exposure of the TEST plates is complete, the TEST plates will be placed into the incubator.
     11. All of the plates will be incubated at 37 degree overnight and examined the following day for growth. The CFU's/ml in the overnight cultures will be
          determined as will the CFU's/ml on the Control and TEST plates.
     12. All results will be recorded by each pair on their data collection sheet.
